Durch Kombination unterschiedlicher natürlicher und nicht natürlicher (auf Tyrosin) basierender Aminosäuren und deren Verknüpfung zu aminoterminal Prolin und Prolinderivate tragenden Peptiden unter Verwendung der Festphasensynthese wurde eine Peptidbibliothek synthetisiert, deren Peptide auf ihre katalytischen Eigenschaften in der Aldolreaktion untersucht wurden. Eine Reihe von Peptiden wurde auch in freier Form in Lösung katalytisch evaluiert. Von Bedeutung sind Umsetzungsgeschwindigkeit der Edukte sowie erzielbare Enantiomerenüberschüsse des Aldolprodukts und Einflüsse von Peptidsequenz, Peptidlänge und aminoterminaler Chiralität der synthetisierten Peptide. Mit Hilfe einer neuen online-NMR-Methode konnte die Kinetik der Aldolreaktion nichtinvasiv beobachtet und so kinetische Konstanten der Aldolkatalyse ermittelt werden. Kraftfeldmethoden, gestützt durch theoretische Überlegungen führen zu Einblicken in die Struktur von Übergangszuständen der Reaktion.
