Description

Aerobic bacteria mostly produce several coloured pigments, based on the location, temperature and other environmental isolated factors from which they get isolated. Present study probes to focus on the production, extraction and applications of the desired pigment-producing bacteria. For this, various soil samples were collected from a nearby local meat shop, followed by their isolation, screening and other succeeding steps. This bacterial blue-green pigment was later found to be pyocyanin, which was isolated from the culture of Pseudomonas aeruginosa. This culture was first isolated and then inoculated in the nutrient broth at an optimal temperature of 37°C for about 48 hours in an orbital shaker. The pigment was extracted by several solvent-extraction methods including solvents such as Chloroform, diethyl ether, Acetone and Ethanol. It was then characterized and analyzed using UV spectrophotometry. Further, the extracted pigment was tested for anti-microbial activity, against the E. coli pathogen and anti-oxidant activity, using a DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. The results were interpreted and analyzed for pyocyanin pigment production and its relevant extraction.
