Das Hauptziel der vorliegenden Untersuchung war die Formulierung und Charakterisierung eines mit β-Cyclodextrin beladenen Indomethacin-Mundlösungsfilms zur Umgehung des First-Pass-Metabolismus Indomethacin ist ein nicht-steroidales entzündungshemmendes Mittel (NSAIA) mit entzündungshemmender, schmerzstillender und fiebersenkender Wirkung. Man nimmt an, dass seine pharmakologische Wirkung durch die Hemmung des Enzyms Cyclooxygenase (COX) vermittelt wird, dem Enzym, das für die Katalyse des geschwindigkeitsbegrenzenden Schritts der Prostaglandinsynthese über den Arachidonsäureweg verantwortlich ist. Die mit Indomethacin-β-Cyclodextrin beladene mundlösliche Folie wurde durch Lösungsmittelverdampfung mit verschiedenen hydrophilen Polymeren wie Hydroxylpropylmethylcellulose (HPMC) 15cps, HPMC K100M, SLS (Natriumlaurylsulfat) und PEG 400 & PEG 600 hergestellt. Die formulierte mundlösliche Folie wurde auf ihre physiochemischen Parameter wie Gewichtsvariation, Dicke, Faltfestigkeit, Arzneimittelgehalt, Feuchtigkeitsgehalt und Feuchtigkeitsabsorption untersucht. Die In-vitro-Wirkstofffreisetzung wurde mit einem Auflösungsgerät durchgeführt. Alle diese mundauflösenden Filme zeigten, dass >70% der Droge innerhalb einer halben Stunde freigesetzt wurden und einer Freisetzungskinetik erster Ordnung gehorchten.
