EGON STAHL Geeignete Trenn- und Nachweisverfahren gehoren beim Arbeiten mit chemischen Stoffgemischen zum Handwerkszeug, und es gibt keine Uni versalmethoden. Liegt ein Vielstoffgemisch vor, so wird man es zunachst mit einer einfachen Methode vortrennell und es beispielsweise in einen lipophilen und einen hydrophilen Anteil zerlegen. Danach werden die beiden Fraktionen weiter aufgeschliisselt, bis man zu Gemischen mit recht ahnlichen Eigenschaften kommt. Zur weiteren Auftrennung der artiger Stoffgruppen verwendet man dann vorteilhaft die Chromato graphie. Mit der Diinnschicht-Chromatographie lassen sich viele Trennpro bleme einfach und schnelllosen. Immer jedoch sollte man sich auch hier dariiber im klaren sein, daB es darauf ankommt, die stationare und die mobile Phase so aufeinander abzustimmen, daB man die optimalen Trennbedingungen erreicht. Die zu beachtenden Zusammenhange zwi schen den drei HauptgroBen der Chromatographie zeigt die Abb.99. Dieses Schema fuBt zunachst auf dem Erfahrungsschatz der Adsorptions Chromatographie; er sei deshalb nachstehend zusammengefaBt: Zu trennendes Gemisch a) Gesiittigte Kohlenwasserstofje werden nicht oder nur gering adsorbiert und wandern daher am schnellsten. Ungcsiittigte Kohlenwasserstofje werden tm so starker adsorbiert, je mehr Doppelbindungen sie enthalten und je mehr davon in Konjugation stehen. Zur Trennung muB man daher ein aktives Adsorptionsmittel und ein wenig polares FlieBmittel verwenden. Eine andere Mtiglichkeit ist die "Phasenumkehr"; die stationare Phase ist hier mit einem Lipid impragniert und als mobile Phase dient ein hydrophiles FlieBmittel. b) TV erden funktionelle Gruppen in einen Kohlenwa8serstofj eingefiihrt, so erhOht sich die Adsorptionsafjinitiit in nachstehender Folge: -CHa-+ -O-Alkyl-+ >C = 0 -+ -NH -+ -OH-+ -COOH."
