In der ersten Halfte unseres Jahrhunderts betrachteten es die Biochemiker als ihre Hauptaufgabe, die Wege des Stoffwechsels in lebenden Zellen auf zuklaren. Heute haben wir ein zwar noch nicht ganz vollstandiges aber doch weitgehend klares Bild von den chemischen Umsetzungen in lebenden Zellen. Etwa 1000 bis 2000 verschiedene Enzyme katalysieren die einzelnen Reak tionsschritte, die zum Auf-, Um- und Abbau von Kohlenhydraten, Pro teinen, Fetten und anderen Stoffklassen fijhren. In den letzten Jahren hat sich das Interesse der Biochemiker bevorzugt der Frage zugewandt, wie die chemischen AbHiufe in lebenden Zellen reguliert werden, urn die fijr alle Lebewesen charakteristische automatische Anpassung des Stoffwechsels an Veranderungen der Umweltbedingungen zu ermoglichen. Schon Anfang der vierziger Jahre haben Fritz Lipmannt, Hermann M. Kalckar2 und Feodor 3 Lynen ein grundlegendes Regulationsprinzip erkannt: Der Phosphatkreis lauf in den lebenden Zellen reguliert den Energiestoffwechsel nach dem Prin zip "die Nachfrage kontrolliert das Angebot". Da einerseits alle energie benotigenden Prozesse der Zelle mit einer Spaltung von ATP (Adenosin triphosphat) zu ADP (Adenosindiphosphat) und Phosphat, bzw. AMP (Adenosinmonophosphat) und Pyrophosphat verbunden sind und anderer seits die energieliefernden Prozesse mit der Synthese von ATP aus AMP bzw. ADP und Phosphat gekoppelt sind, kann der ATP-liefernde Energie stoffwechsel nur in dem Urn fang ablaufen, wie vorher ATP verbraucht Tab.
