I wrote this book based on a 3-credit hour graduate course, "Advanced Pharmacokinetics and Pharmacodynamics", that I have been teaching in the past 10 years. The content of my course was based on my two-decade-long PK/PD research experience plus knowledge learned from multiple PK/PD courses from SUNY, University at Buffalo, whose PK/PD program has been #1 in the world for several decades. If you want to be a pharmacometrician and/or become a modeling expert, this book is for you. The first half of the book (Chapters 1-7) teaches the fundamentals, including how to draw model diagrams, how to write PK differential equations, and how to solve them using Laplace transformation. The second half of the book (Chapters 8-16) covers more advanced topics, including various PK models (e.g., nonlinear PK, TMDD model, whole-body PBPK, PopPK), PD models (e.g., effect compartment, indirect response models) and QSP models (e.g., platform QSP models for new drug modalities). Appendix 1 shares the scientific journey of a legend in PK/PD- interview with Dr. William Jusko. In addition, I included the history of many important models in this book so that readers could get a big picture of the PK/PD discipline and have a deeper understanding of “where we came from” and “where we are now”. Note: This book targets advanced users and is not suitable for beginners who just stepped into the PK/PD world. For beginners, please consider reading my basic PK textbook ("Essentials in Clinical Pharmacokinetics: Concepts, Dose Optimization, and Biologics", which is also available at Amazon) to build a solid foundation first before reading this book.
