LIPOGENESE DER ARTERIENWAND: REGULIERUNG IHRER EXPRESSION UND ANOMALIEN BEI INSULINRESISTENZ UND DIABETES Wir haben die Expression und Regulation der Lipogenese in Aorten und CMLV untersucht und festgestellt, ob sie durch Insulinresistenz und Diabetes verändert wird. Fettleibige Zucker-Ratten (ZO), diabetische Ratten und Psammomys obesus-Ratten lagern mehr Lipide in ihren Aorten an als ihre Kontrolltiere. Die Expression der Gene für die Lipogenese und der Gene, die an der Fettsäureaufnahme beteiligt sind, ist jedoch nicht erhöht. Ein ähnliches Ergebnis wurde bei Endarteriektomien beim Menschen festgestellt. Das adipogene Milieu, Glukose oder Insulin allein stimulieren die Lipogenese nur in den CML von Zucker-Kontrolltieren moderat, in den CML von ZO wurde keine Wirkung beobachtet. Wir haben gezeigt, dass die Auswirkungen von TO901317 auf die Lipogenese in CMLV ausschließlich auf die Aktivierung des nukleären Rezeptors LXRα zurückzuführen sind, PXR hat keine Wirkung. Zusammenfassend lässt sich sagen, dass die Lipogenese in der Arterienwand während Insulinresistenz und Diabetes nicht erhöht ist.
