Milchsäurebakterien (LAB) sind aufgrund ihrer fermentativen Fähigkeit wirtschaftlich wichtig und bieten auch gesundheitliche und ernährungsphysiologische Vorteile. Pediococcus acidilactici NCDC 252 (NCDC 252) ist ein potenzielles Probiotikum mit neuartigem Genom, das alle probiotischen In-vitro-Eigenschaften, die Kolonisierung im Darm und das Überleben unter Bedingungen aufweist, die der Magen-, Darm- und Vaginalflüssigkeit ähneln. Das Genom wurde auf seine Fähigkeit untersucht, antimikrobielle Peptide (AMP) zu produzieren. AMP aus NCDC 252 wurde isoliert und gegen den Indikatorstamm (Escherichia coli) gereinigt. Die AMP-Produktion wurde in MRS-Brühe bei 32 °C, pH 6 und einer Inkubationszeit von bis zu 36 Stunden unter aeroben und anaeroben Bedingungen optimiert. AMP wurde 2,2-fach mit einer Ausbeute von 7,2 % und einer spezifischen Aktivität von 64,5 U mg/ml mittels Ammoniumsulfatfällung (bis zu 70 %) und Gelfiltrationschromatographie gereinigt. Das Molekulargewicht des gereinigten AMP wurde mit 2,5 KDa berechnet. Tricin-SDS-PAGE- und LC/MS-Studien bestätigten, dass es sich um ein Peptid mit niedrigem Molekulargewicht handelt. AMP war im pH-Bereich von 3 bis 9 und im Temperaturbereich von 40 °C bis 100 °C stabil. Die enzymatische Behandlung inaktivierte das AMP, wohingegen die Behandlung mit Detergenzien seine Aktivität steigerte.
