Description

Parodontitis wurde bei männlichen Wistar-Ratten durch Injektion von E.coli-Endotoxin in die labialen und palatinalen Bereiche der vorderen Oberkieferzahnhälse sowie die bukkalen und palatinalen Bereiche der Oberkiefermolaren ausgelöst. Die Proanthocyanidine (PC) werden aus Traubenkernen extrahiert. Die schützende Wirkung verschiedener PC-Dosen wurde im Blut untersucht, indem die reaktiven Sauerstoffspezies, die lysosomalen Enzymaktivitäten sowie nicht-enzymatische und enzymatische Antioxidantien bestimmt wurden. Der histopathologische Nachweis einer experimentellen Parodontitis zeigte eine zelluläre Infiltration von Entzündungszellen, während die mit Proanthocyanidin behandelten Gruppen nur vereinzelte Entzündungszellen und Blutgefäße aufwiesen. Histopathologische und immunhistochemische Untersuchungen des Lebergewebes zeigten, dass die langfristige Supplementierung von PC keine toxische Wirkung hatte. Die durch Endotoxin induzierte Parodontitis wurde klinisch durch die Bildung von Zahnfleischtaschen beurteilt und korrelierte mit histopathologischen und biochemischen Untersuchungen. Die Rolle von PC wurde in vitro bei der Modulation der Neutrophilen- und Makrophagenfunktion untersucht.
