Mitochondriën zijn de plaats van oxidatieve fosforylering in alle eukaryote cellen en zijn de voornaamste centra van ATP-productie. Plantenmitochondriën hebben een sterk vertakte ETC die flexibiliteit biedt voor de oxidatie van cytosolisch en matrix NAD(P)H. Naast de universele cytochroomroute die in veel organismen wordt aangetroffen, hebben planten matrix-georiënteerde en cytosol-georiënteerde niet-fosforylerende NAD(P)H-dehydrogenases die Complex I omzeilen en het matrix-georiënteerde Alternatieve oxidase (AOX) dat de Complexen III en IV omzeilt. De alternatieve routes zijn niet-proton pompend en maken NAD(P)H oxidatie mogelijk met verschillende niveaus van energiebehoud en daarom is deze alternatieve route van bijzonder belang. Recente studies hebben aangetoond dat de alternatieve ademhalingswegen worden gestimuleerd tijdens fysiologische stress en er wordt gesuggereerd dat deze alternatieve ademhalingswegen adaptieve kenmerken van planten vertegenwoordigen. De doelstellingen van het project omvatten de creatie van transgene A.thaliana planten met verhoogde niveaus, door overexpressie, en verlaagde niveaus, door RNAi, van het cytosol gerichte niet-fosforylerende NAD(P)H-dehydrogenase Atndb2 en analyse van het effect van zoutstress op transgene A.thaliana waarin AOX is uitgeschakeld.
