Le travail présenté dans ce mémoire concerne la synthèse d'aminoacides silylés et l'incorporation dans des composés biologiquement actifs tels que la substance P, la bradykinine, la neurotensine ou le captopril. L'importance des aminoacides non- protéinogéniques n'est plus à démontrer depuis quelques années en raison de leur non-reconnaissance par les systèmes naturels de dégradation protéolytique. La proline, ainsi que ces analogues, intervient directement dans la structuration tridimensionnelle des peptides. Ces structures peuvent également être induites par des aminoacides dialkylés non-codés. Le silicium accroît la lipophilie des aminoacides silylés d'où une meilleure pénétration des membranes cellulaires des peptides-hôtes ainsi qu'une augmentation des interactions hydrophobes. Le texte s'articule autour de trois points: synthése de nouveaux aminoacides silylés, mise au point d'une nouvelle méthodologie de synthèse, synthése de peptides biologiquement actifs incorporant la triméthylsilylalanine et la silaproline. Ces analogues ont été testés en terme de résistance enzymatique, d'affinité et de reconnaissance récepteur-ligand.
