Depuis l?invention du laser en 1960, ce dernier a beaucoup évolué, et s?est diversifié jusqu?à s?implanter sérieusement dans notre vie quotidienne. Mais les lasers restent avant tout des outils formidables pour les scientifiques. Dans cette thèse est présentée une source laser novatrice compacte, un Laser à Electrons Libres (LEL) dit « injecté ». Alors que les sources conventionnelles de LEL, basées sur l?émission spontanée auto-amplifiée, possèdent une cohérence temporelle partielle, nous démontrons ici la forte amplification cohérente de la 5ème harmonique d?un laser Ti: Sa générée dans une cellule de gaz (160 nm), puis injectée dans le LEL du prototype de l?accélérateur SCSS au Japon. Vu le faible niveau d?injection requis, des rayonnements totalement cohérents pourraient être générés jusqu?à la "fenêtre de l?eau", zone spectrale pour laquelle le rayonnement laser est non absorbé dans l?eau et donc dans les tissus biologiques. Cela permettrait ainsi de sonder précisément la matière pour observer les principaux phénomènes physiques et chimiques qui se déroulent sur une échelle temporelle ultra-rapide.
