Pour avoir employé, à côté de la doxologie traditionnelle "Gloire au Père, par" le Fils, "dans" le Saint-Esprit" , la doxologie "Gloire au Père, "avec" le Fils, "avec" le Saint-Esprit" , Basile s'était vu accuser d'utiliser deux formules contradictoires. En réalité, la seconde privait à la fois les ariens et les pneumatomaques de l'argument qu'ils tiraient des prépositions utilisées dans la doxologie traditionnelle pour établir l'infériorité du Fils et de l'Esprit par rapport au Père. A l'origine de l'ouvrage, il y aurait donc, de la part de Basile, un désir de justification. Amphiloque d'Iconium, qui l'a interrogé sur la signification de ces particules grammaticales, lui en fournit le prétexte. Toutefois, l'enseignement sur le Saint-Esprit délivré par Basile, en 375, pour répondre à son attente, est de toute évidence le fruit d'une longue maturation. Toute sa démonstration vise à établir l' "homotimie" - l'égalité d'honneur -, de l'Esprit avec le Père et avec le Fils, à partir de l'Ecriture et de la Tradition. L'examen attentif des particules dans chaque formule étudiée, à commencer par la formule baptismale, confirme cette "homotimie" du Fils avec le Père, et de l'Esprit avec le Père et le Fils. Utiliser "avec" dans la doxologie, pour glorifier l'Esprit, est donc parfaitement légitime. Jamais pourtant, la divinité ou la consubstantialité de l'Esprit ne sont ouvertement affirmées par Basile. Ce silence, à première vue surprenant, au regard de toute la démonstration, est sans doute "politique" . L'introduction par Basile du terme "homoousios" - il l'évite ici, même pour le Fils - aurait risqué d'accroître encore les divisions dans l'Eglise, s'il l'avait appliqué au Saint-Esprit. Son traité n'en a pas moins influencé profondément la réflexion théologique postérieure, en Orient comme en Occident. Entre les conciles de Nicée (325) et de Constantinople (381), il marque une étape décisive dans l'élaboration de la théologie du Saint-Esprit".
